Hamilton was educated at Glasgow University before moving to Ireland to manage his uncle's estate. While there, he was influenced by the Presbyterian preacher Robert Blair and was ordained himself in 1625, becoming minister at Ballywalter, county Down. He was deposed and returned to Scotland in 1637, where he became minister at Dumfries before moving to Edinburgh in 1645. After he was deprived in 1660, he went to Inveresk.
